The Intelligence Dampening Sphere, more commonly known as Wheatley, is a personality core of masculine programming and English West Country accent who appears in the single-player campaign of Portal 2. He serves as the game's deuteragonist, guiding Chell through her efforts in escaping the Aperture Science Enrichment Center. However, half-way through the game he and GLaDOS switch roles as she becomes Chell's deuteragonist and Wheatley becomes the game's main antagonist. Wheatley is voiced by British actor and comedian Stephen Merchant.

The Adventure Sphere (who calls himself "Rick") is a corrupted personality core of male programming who appears in the single-player campaign of Portal 2. He is one of the cores retrieved by GLaDOS for Chell to attach onto Wheatley, in attempt to initiate a core transfer procedure via corrupting his control of the central core body. The Adventure Sphere speaks in the manner of a stereotypical "tough guy", spending most of his time boasting about his bravery and excitement for danger. Upon seeing Chell, he flirts with her and makes increasingly chauvinistic comments. He even tries (but to no avail) to convince Chell to take a "lady break" while he deals with Wheatley by himself. Fact is one of the cores retrieved by GLaDOS for Chell to attach onto Wheatley, in order to initiate a core transfer procedure via corrupting his control of the central core body. The Fact Sphere speaks like a stereotypical "know-it-all", spending most of its time rapidly rattling off various facts. While a small portion of the listed facts are true, most of them are incorrect and downright illogical. (Sometimes altering true facts into nonsense) The Fact Sphere, along with all the corrupted cores, is voiced by Nolan North.

The Morality Core is a personality core that appears in Portal, and Portal 2: Lab Rat. It is one of many cores that was attached to GLaDOS in order to regulate her behavior. It was programmed to prevent her from doing unethical deeds, acting as an artificial conscience. When Doug Rattmann first sees the Morality Core, he doubts its capability to restrain GLaDOS, saying "You can always ignore your conscience". He turns out to be correct, as despite the Morality Core being attached, GLaDOS floods Aperture with neurotoxin anyway. At the end of Portal, it falls off of GLaDOS' body, possibly intentionally. GLaDOS repeatedly tells Chell not to touch it, but she disregards this advice and destroys it in an Emergency Intelligence Incinerator. When incinerated, it is then known by GLaDOS as the "Aperture Science Thing We Don't Know What It Does". With her conscience removed, GLaDOS is free to flood her chamber with neurotoxin

The Curiosity Core is a personality core that appears in Portal. It is one of the cores Chell knocks off of GLaDOS during the climactic "battle" at the end of the game and later destroys in an Emergency Intelligence Incinerator. The Curiosity Core is an inquisitive, active chatterbox that speaks with a high-pitched, almost child-like, voice. The Curiosity Core is voiced by Ellen McLain

The Intelligence Core, also referred to as the sphere_cakemix in the game files, is a personality core that appears in Portal. It is one of the cores Chell knocks off of GLaDOS during the climactic "battle" at the end of the game. It was later destroyed in an Emergency Intelligence Incinerator. The Intelligence Core speaks in a slow, monotonous voice, and spends its time reciting a cake recipe, which often has unusual or potentially lethal ingredients. As it speaks, it will roll lazily around, seemingly entranced in recipe recitation. The Intelligence Core is voiced by Ellen McLain. The recipe he gives in LEGO Dimensions consists of "fish-shaped solid waste, a single banana, and a cup of lemonade." These are all included in the actual cake recipe.

The Anger Core is a personality core that appears in Portal. It is one of the cores Chell knocks off of GLaDOS during the climactic "battle" at the end of the game. It was later destroyed in an Emergency Intelligence Incinerator. The Anger Core is voiced by Mike Patton.

The Party Escort Bot or Party Associate, is a mysterious pink-eyed core and android that was originally cut from Portal until it was later added into the game's ending as part of the March 3, 2010 patch

Defective Turrets, also called "Crap Turrets" by Wheatley, are damaged, ammo-less, unfinished, or faultily assembled Sentry Turrets. Unlike normal Sentry Turrets, Defective Turrets have a more humorous and sarcastic dialogue, and speak in a masculine, less robotic voice. They are harmless to Chell, many times making comments such as "If anyone asks, I killed you". Their voice lines seem to imply they are blind, but they are still aware of Chell's presence. They are first encountered in Chapter 5: The Escape. The Defective Turrets are voiced by Nolan North.

Turrets serve as one of the main testing obstacles in the Portal series. Armed with almost unlimited ammunition and deadly accuracy, they will attempt to kill test subjects on sight. Standard Sentry Turrets are voiced by Ellen McLain, while Defective Turrets are voiced by Nolan North.

GLaDOS (Genetic Lifeform and Disk Operating System) is the central core designed to control, guide, and oversee the Aperture Science computer-aided Enrichment Center. With the ability to shift and move facilities and various chambers, GLaDOS is able to achieve a seamless and almost infinite testing design whilst residing in the Central AI Chamber. In possession of a feminine programming, she is the antagonist of Portal and the first half of the single-player campaign in Portal 2. During the later half, she becomes the game's tritagonist. In the game's cooperative campaign, GLaDOS is the testing supervisor of Atlas and P-body. GLaDOS is voiced by opera singer and voice actress Ellen McLain
