The one you first start off with is what I use for drawing main things and bases
The paint roller is used for shading, blending and making colours
The fill bucket is for, well.. filling
The little ink dropper is for picking up colours
The chisele can only draw on colour, not the normal grid
The highliter can be used for shading
The eraser is for erasing oWo
And the pencil is for if you want smooth lines
